He returned to Ottoman Empire in 1914 to be general director of all Armenian schools in Moush region. Prepared Armenians for self-defense after Ottoman Empire entered the first world war. Ruben Ter Minassian played a key role in the revolution movement of Kars, Van and other places, where there was happening Genocide by the Ottoman Empire during World War I. Resisted the Ottoman troops for several months in 1915, breaking through a circle of them. In 1915, he led the defense of the Armenians of Sassun against the Third Army. He was a member of the Armenian National Council in 1917. An adviser to the Transcaucasian Sejm delegation in the Trebizond negotiations with the Ottoman Empire in March 1918.
