• Lara Croft: The young adventuress starts her journey here, when she is shipwrecked on an island populated with dark secrets, vicious animals and mercenaries who seem intent on killing her and the other survivors.
• Conrad Roth: Captain of the Endurance, the ship on which Lara travels when it is struck by a storm and splits the ship in two. It has been stated that he will act as a mentor figure for Lara through the course of the game.
• Sam: Lara's best friend and fellow graduate who is also the crew's camerawoman. The mysteries surrounding the island have some connection with Sam when she is kidnapped by the islanders.
• Mathias: The leader of the Solarii cult on the island and the main antagonist of the game. Marooned on the island for decades, he plans to sacrifice Sam so that he can finally escape the island.
• Dr. James Whitman: A celebrity archaeologist who Lara once admired, and who would do anything to fulfil his ambitions of fame and fortune.
• Reyes: Mechanic of the Endurance, who gets along very well with Roth but is rather hard on Lara due to her inexperience.
• Jonah: The easy-going fisherman who believes in supernatural events such as the curse of the island.
• Grim: The old helmsman of the Endurance who had worked alongside Roth and Lara's father for many years.
• Alex: A computer specialist who harbors feelings for Lara. He hates walls.
• Himiko: The deceased Sun Queen who once ruled over the island of Yamatai and at the center of the island's secrets.
