Tachykinins selectively bind and activate the G-protein coupled receptors NK1R, NK2R, and NK3R. Neurokinin A binds to the G-protein coupled receptor ultimately increasing the release of inositol-phosphate and calcium second messengers. Each receptor demonstrates a specific affinity for either neurkinin A or substance P peptides. Both peptides, however, can act as full agonists on either receptor, although their potency is decreased when not bound to their specific receptor.
