Nescopeck Creek is born in Hanover Township, Luzerne County, Pennsylvania at the confluence of an outlet of Olympus Pond and Creasy Creek. Nescopeck Creek flows southwest for several miles through Nescopeck State Park. The creek then continues flowing southwest, across Pennsylvania Route 309, whereupon the creek starts flowing west, roughly paralleling Interstate 80 and picking up Little Nescopeck Creek. A few miles further on, Nescopeck Creek turns northwards, picks up Black Creek, and crosses Interstate 80. The creek then flows northwest, making several meanders, and emptying into the Susquehanna River in Nescopeck.
