Jamestown High School offers a huge selection of sports including Softball, Baseball, Boys Tennis, Boys and Girls Track and Field, and Golf in the spring. Football, Boys and Girls Soccer, Boys and Girls Cross Country, Girls Tennis, Girls Swimming, and Girls Volleyball in the fall. Boys and Girls Basketball, Boys Swimming, Boys and Girls Indoor Track, Bowling, Wrestling and Hockey in the winter. Hockey is not part of the school's varsity sports program but is run by the local youth hockey association in Jamestown. The school's mascot is a cartoon Native American man carrying a tomahawk.

In 2011, thanks mainly to senior forward Darin Butts and junior standout Jaysean Paige, the Jamestown Red Raiders were runner- ups for the State in basketball, eventually falling to state/national powerhouse Mt. vernon. the basketball squad absolutely dominated Section 6 and won against some of the state's top teams, including Niagara Falls and Albany CBA.
