- Michael Weatherly as Anthony "Tony" DiNozzo- Gibbs's second in command and also the Senior Special Agent of the team.
- David McCallum as Dr. Donald "Ducky" Mallard- NCIS's Medical Examiner.
- Pauley Perrette as Abigail "Abby" Sciuto- NCIS's Forensic Scientist who wears Goth-style clothing.
- Robyn Lively as Special Agent Vivian Blackadder – A former FBI Agent who lost her brother in the USS Cole bombing, prompting her to join NCIS where she became a member of Gibb's team as she sought to avenge her brother's death. She along with Don Dobbs are the only two who appeared in the following episode of JAG, but did not appear in NCIS itself.
- Thomas Anthony Jones (aka Tom Jay Jones) as Special Agent Don Dobbs- An NCIS Special Agent who helps the team with the investigation into Singer's murder but like Blackadder, Dobbs did not appear in the main NCIS TV series.
