Eastern's sports teams are known as the Quakers and are members of the Capital Area Activities Conference, with intracity rivals Everett High School and Sexton High School.

The Don Johnson Field House is located behind Eastern. Inside, the Field House contains a basketball court named for Paul Cook, a former Quaker Men's Basketball coach and the all-time winningest coach in Michigan High School Athletic Association history. It is big enough to seat the entire student body inside on the bleachers.

As of 2011, the Eastern wrestling team had the highest G.P.A. of any wrestling team in the region.

Lansing Eastern is one of the few high schools in the area with a competitive Winter Guard. The guard was established in the early 2000s. At a recent competition the guard earned enough points to be boosted up a division and is now at the AAA level.
