- Ajay Devgn as Karan, alias Doc. The main protagonist of the film, and also referred to as the mastermind of thiefs. He hides his criminal life from everyone including his girlfriend, Shania.
- Sunil Shetty as Angad; the main antagonist of the film. Angad, who has been attacked several times by friends, family members, and less enemies, has always survived the bullets. The story focuses on his intentions to steal the diamonds the most.
- Ritesh Deshmukh as Lucky; an small-time thief who wants to make big like Karan. Lucky is noted to dislike Dhananjay, but throughout the film, the two become friends.
- Zayed Khan as Dhananjay Jhumbevalkar, alias Danny or simply DJ. Another small thief with a big mind, who also dislikes Lucky.
- Dia Mirza as Aditi; another supporting antagonist in the film. She is Angad's girlfriend, but does not know that Angad wants the diamonds for himself.
- Esha Deol as Puja; Lucky & Dhananjay's love interest.
- Shamita Shetty as Shania; Karan's long-time girlfriend. An undercover spy.
- Ayesha Takia as Preeti/Ria; an fellow passenger of Dhananjay's. She is also part of the main story, however her real character is revealed throughout the end of the film.
